Rune Tablets are interactive objects found at several locations across The Sea of Thieves.
The tablets were a utilized as part the teasers for Season Three that spanned through game updates and cryptic social media posts.
Lore
Duke spent a while scouring islands during an investigation into current events. As part of this investigation he found tablets with Skeleton Runes that he'd try to translate, leaving behind notes for possible translations.
There were also a handful of tablets that could be found which Duke presumably did not discover himself as he never mention these locations and there are no translations left among them.

Location: Found on the rock ledge above the waterfall nearest the cave.
This location closely matches the area where YouTuber Captain Falcore first revealed his "Ghost of Crook's Hollow" hoax.
